Genetic Upregulation of Aromatase via Inflammatory Signaling

The human aromatase gene has several tissue-specific promoters; in adipose tissue, its expression is primarily driven by promoter I.4, which is highly responsive to inflammatory signals. The binding of NF-κB, along with the activation of other signaling pathways stimulated by cytokines, effectively “turns on” the gene, leading to a marked increase in the transcription of aromatase mRNA and, consequently, the synthesis of the aromatase enzyme itself. This creates a direct, mechanistic link ∞ adipocyte death recruits macrophages, which release cytokines, which activate NF-κB, which drives aromatase gene expression.
