Tight Junction Modulation is the physiological or therapeutic manipulation of the paracellular permeability of epithelial and endothelial barriers, particularly in the gastrointestinal tract and the blood-brain barrier. Tight junctions are multi-protein complexes that seal the space between adjacent cells, controlling the passage of molecules. Modulation refers to the clinical effort to restore compromised barrier function, often termed “leaky gut,” which is implicated in systemic inflammation and autoimmune conditions. This restoration is crucial for preventing the entry of inflammatory antigens and supporting immune homeostasis.
