The HPT axis is a classic model in endocrinology, with the regulatory loop being fully described in the mid-20th century following the isolation and characterization of Thyrotropin-Releasing Hormone (TRH) and Thyroid-Stimulating Hormone (TSH). The term ‘regulation’ emphasizes the dynamic, self-adjusting nature of this system, which constantly monitors and adjusts hormone output to meet the body’s fluctuating metabolic demands. Mechanism
Regulation begins with the hypothalamus releasing TRH, which stimulates the anterior pituitary to secrete TSH. TSH then acts on the thyroid gland to produce thyroxine (T4) and triiodothyronine (T3). Crucially, T4 and T3 exert negative feedback on both the hypothalamus and pituitary, slowing the release of TRH and TSH when levels are sufficient. Peripheral conversion of the prohormone T4 to the more active T3 is also a major regulatory point, influencing the hormone’s final biological impact on target tissues.
