This term is a foundational concept in endocrinology, describing the hierarchical control system that links the central nervous system to the thyroid gland. Its understanding is rooted in classical physiological studies that identified the roles of Thyrotropin-Releasing Hormone (TRH) from the hypothalamus and Thyroid-Stimulating Hormone (TSH) from the pituitary. The axis model is critical for diagnosing and managing thyroid disorders.
Mechanism
The hypothalamus releases TRH, which stimulates the pituitary to release TSH. TSH then acts on the thyroid gland to stimulate the production and release of T4 and T3. These thyroid hormones circulate and exert negative feedback on both the pituitary and the hypothalamus, tightly regulating their own concentration. The peripheral conversion of T4 to the more active T3, and the subsequent binding of T3 to nuclear receptors, is the final effector mechanism that drives changes in gene expression and metabolic rate across target tissues.
