Thermal denaturation is the irreversible physical process by which the complex, highly specific three-dimensional structure of a biological macromolecule, such as a protein, enzyme, or hormone receptor, is fundamentally disrupted by exposure to excessive heat. This loss of the native, functional conformation results in the complete loss of the molecule’s biological activity, such as an enzyme losing its catalytic function or a hormone-binding protein losing its capacity to transport. This principle is clinically relevant in ensuring the integrity of biological samples for accurate hormonal analysis.
