A suicide inhibitor is an irreversible enzyme inhibitor that binds to an enzyme’s active site and is then catalytically modified by the enzyme itself. This transformation converts the inhibitor into a highly reactive molecule, forming a stable, covalent bond with a critical residue within the active site, permanently deactivating the enzyme. The term “suicide” reflects the enzyme’s role in its own inactivation.
