A Senolytic Intervention is a clinical or pharmacological strategy specifically designed to selectively induce apoptosis, or programmed cell death, in senescent cells—cells that have ceased dividing but remain metabolically active and secrete pro-inflammatory molecules. These senescent cells accumulate with age and contribute significantly to chronic inflammation, tissue dysfunction, and the pathology of numerous age-related diseases. The goal of this intervention is to clear this toxic cellular burden, thereby rejuvenating tissue microenvironments and improving organ function.
Origin
The term “senolytic” is a combination of “senescence” and “-lytic” (meaning to destroy), coined following the discovery of compounds that could selectively eliminate senescent cells in culture. This concept is a core pillar of modern longevity science, offering a direct therapeutic avenue to address one of the fundamental hallmarks of biological aging.
Mechanism
The mechanism relies on exploiting the unique pro-survival pathways that senescent cells activate to resist their own apoptosis, known as the Senescence-Associated Anti-Apoptotic Pathways (SAAPs). Senolytic agents work by disrupting these SAAPs, often targeting proteins like Bcl-2 or PI3K/AKT, thereby making the senescent cells vulnerable to their own internal death signals. The selective clearance of these dysfunctional cells reduces the Senescence-Associated Secretory Phenotype (SASP), mitigating chronic systemic inflammation and promoting tissue repair.
