The biological process of clinically or physiologically regulating the cellular senescence pathway, which is the state of irreversible cell cycle arrest that older or damaged cells enter. Modulation aims to clear senescent cells, often called “zombie cells,” or to mitigate the pro-inflammatory secretome they release, known as the Senescence-Associated Secretory Phenotype (SASP). This intervention is a central strategy in anti-aging and longevity medicine to reduce tissue dysfunction.
Origin
The term originates from the field of gerontology and molecular biology, where the senescence pathway was first characterized as a tumor-suppressive mechanism. Its application to systemic health and longevity, or ‘modulation,’ arose from the discovery that the accumulation of these cells contributes significantly to age-related inflammation and chronic disease. Endocrinology links this to age-related hormonal decline, which can exacerbate SASP effects.
Mechanism
Modulation is primarily achieved through senolytic agents, which selectively induce apoptosis in senescent cells, or senomorphic agents, which suppress the harmful SASP. By reducing the burden of senescent cells and their inflammatory output, tissue function is improved, and chronic, low-grade systemic inflammation is decreased. This reduction in inflammatory signaling helps to restore hormonal sensitivity and metabolic efficiency across various organs.
