Receptor Affinity Optimization is a therapeutic goal aimed at enhancing the sensitivity and binding strength of cellular receptors to their specific signaling molecules, such as hormones, growth factors, or neurotransmitters. This strategy is critical when circulating hormone levels are adequate but the biological response is attenuated, often due to downregulation or desensitization of the receptors. Optimizing affinity allows for a maximal biological effect from a normal physiological concentration of the ligand.
