RANK/RANKL Pathway

Meaning

The RANK/RANKL pathway is a crucial molecular signaling cascade that primarily regulates bone remodeling, but its influence extends to immune function and hormonal regulation of mammary gland development. Receptor Activator of Nuclear factor kappa-B (RANK) is the receptor expressed on the surface of pre-osteoclasts and dendritic cells, while its ligand (RANKL) is expressed on osteoblasts and T-lymphocytes. The binding of RANKL to RANK is the essential signal for osteoclast differentiation, activation, and survival, making this pathway central to maintaining skeletal integrity and responding to hormone-mediated bone loss.
