Prior Approval Supplement

Meaning

A Prior Approval Supplement, often abbreviated as PAS, is a formal regulatory application submitted to the Food and Drug Administration (FDA) to request permission for a major change to an already approved New Drug Application (NDA) or Biologics License Application (BLA). This submission is mandatory for any proposed modification deemed to have a substantial potential to negatively affect the drug product’s identity, strength, quality, purity, or potency. It represents the highest level of regulatory scrutiny for post-approval changes.
