A Pit-1 mutation is a genetic alteration in the POU1F1 gene, encoding the Pit-1 transcription factor. This protein is essential for anterior pituitary development, differentiating cells producing growth hormone, prolactin, and thyroid-stimulating hormone. Such mutations disrupt normal hormone production, leading to combined pituitary hormone deficiencies impacting growth and metabolism.
