Origin
The PI3K enzyme was first identified and characterized by researchers in the late 1980s, and subsequent research rapidly established its role as a key, non-negotiable component in cellular signal transduction. The term ‘signaling’ describes the relay of crucial information from the cell surface receptor down to the nucleus, a process vital for coordinating cellular response to external stimuli. Its clinical importance grew exponentially as its central role in cell survival and anabolic metabolism became unequivocally clear.
Mechanism
The pathway is activated when an extracellular ligand, such as insulin or IGF-1, binds to its specific receptor tyrosine kinase (RTK), leading to the phosphorylation of the receptor’s intracellular domain. This phosphorylation recruits and activates PI3K, which then phosphorylates the membrane lipid PIP2 to generate the critical second messenger PIP3. PIP3 then recruits and activates the protein kinase Akt, which subsequently phosphorylates numerous downstream targets, ultimately promoting anabolic gene expression and inhibiting the cellular process of apoptosis.
