Phase I Hydroxylation is the initial, crucial step in the liver’s two-phase system for metabolizing and preparing lipophilic compounds, including steroid hormones and various toxins, for excretion. This biochemical reaction primarily involves the Cytochrome P450 (CYP450) enzyme superfamily, which introduces or exposes a reactive functional group, most commonly a hydroxyl group (-OH), onto the substrate molecule. This modification increases the compound’s polarity, making it slightly more water-soluble and creating a suitable site for the subsequent conjugation reactions of Phase II. The efficiency of this phase is vital for hormonal clearance and managing the body’s overall toxic burden.
