Pharmacodynamic Unpredictability

Meaning

The inherent clinical uncertainty regarding the magnitude and nature of a drug’s biological effect on the body, particularly when the drug is non-standardized, compounded, or administered outside of its validated therapeutic window. This unpredictability stems from variations in receptor binding, post-receptor signaling, and downstream physiological responses that cannot be reliably anticipated or controlled. In hormonal therapy, it means the clinical outcome of a given dose is not consistently reproducible across all individuals or even within the same individual over time.
