OPG/RANKL Axis

Meaning

The OPG/RANKL Axis is a critical molecular signaling pathway that governs the delicate balance between bone formation and bone resorption, fundamentally regulating bone mineral density and skeletal integrity. It involves three key proteins: Osteoprotegerin (OPG), Receptor Activator of Nuclear factor Kappa-B Ligand (RANKL), and the RANK receptor. In hormonal health, this axis is highly sensitive to sex steroids and parathyroid hormone, making it a central mechanism in post-menopausal osteoporosis and other endocrine-related bone disorders. Maintaining the appropriate ratio of OPG to RANKL is essential for preventing pathological bone loss.
