Origin
Nrf2 was discovered as a transcription factor in the early 1990s, with its critical role in cellular defense elucidated through molecular biology and toxicology studies. The protein is named for its homology to other nuclear factors involved in erythroid gene expression. Mechanism
Under normal conditions, Nrf2 is sequestered in the cytoplasm by its inhibitory protein, Keap1, which targets it for degradation. Upon exposure to oxidative stress or specific phytochemical activators, Keap1 undergoes a conformational change, liberating Nrf2. The freed Nrf2 then translocates to the cell nucleus, where it binds to the Antioxidant Response Element (ARE) sequences in the promoter regions of target genes. This binding upregulates the expression of antioxidant enzymes, effectively increasing the cell’s capacity to neutralize free radicals and clear toxic metabolites. Furthermore, this activation is hormonally regulated, with estradiol promoting its function and testosterone tending to suppress it.
