The term is a compound of “neuro,” referring to the nervous system, “steroid,” indicating the class of lipid-soluble hormones, and “genesis,” meaning creation or origin. The concept was formally established in the 1980s when scientists demonstrated that brain cells possessed the necessary enzymatic machinery to produce steroids directly from cholesterol. This discovery fundamentally expanded the understanding of steroid action beyond the traditional endocrine signaling model.
Mechanism
The process begins with cholesterol, which is transported into the mitochondria of neurons and glial cells, the supporting cells of the nervous system. The enzyme P450 side-chain cleavage (P450scc) converts cholesterol to pregnenolone, the foundational precursor for all steroids. Subsequent localized enzymatic transformations convert pregnenolone into various neuroactive steroids. These neurosteroids often act rapidly by modulating the function of key neurotransmitter receptors, such as GABA-A and NMDA receptors, directly influencing neural communication speed and intensity.
