Mechanism
NPY is co-expressed with Agouti-related peptide (AgRP) in a specific set of neurons within the arcuate nucleus of the hypothalamus. These neurons are tonically inhibited by long-term satiety signals, such as leptin and insulin. When energy stores are depleted, or ghrelin levels rise, this inhibition is lifted, leading to increased NPY synthesis and release. NPY then binds to Y receptors in various hypothalamic nuclei, initiating the cascade that stimulates intense hunger and drives the seeking of food, particularly high-calorie options.
