Mechanism
Mitophagy is typically initiated when a mitochondrion is depolarized, leading to the accumulation of the protein PINK1 on its outer membrane. PINK1 then recruits and activates the E3 ubiquitin ligase Parkin, which tags the damaged mitochondrion for engulfment by an autophagosome. Enhancing this pathway often involves compounds that mimic cellular stress or directly activate the PINK1-Parkin signaling cascade, promoting clearance and renewal of the mitochondrial population.
