Melanocortin Receptor Desensitization is the cellular phenomenon of a transient or sustained reduction in the responsiveness of melanocortin receptors (MCRs) to their endogenous ligands, such as alpha-Melanocyte-Stimulating Hormone (α-MSH), or to synthetic agonists. This process occurs following prolonged or intense receptor stimulation, leading to a noticeable decrease in the magnitude of the downstream intracellular signaling cascade. Clinically, this desensitization can result in a diminished therapeutic effect over time, often necessitating careful adjustments in dosing or a temporary pause in the treatment strategy.
