MC2R stands for Melanocortin 2 Receptor, a specific G-protein coupled receptor predominantly expressed in the cells of the adrenal cortex. This receptor is the primary binding site for Adrenocorticotropic Hormone (ACTH), the pituitary hormone that stimulates the adrenal glands to synthesize and secrete cortisol and other adrenal steroids. MC2R activation is the crucial initial step in the body’s physiological response to stress, initiating the Hypothalamic-Pituitary-Adrenal (HPA) axis cascade. A functional MC2R is essential for maintaining adequate cortisol production, which is vital for glucose metabolism, immune regulation, and blood pressure control.
