Hormonal balance, particularly optimal levels of estrogen and testosterone, significantly influences the induction and maintenance of LTP, underscoring the endocrine system’s role in cognitive health.
Origin
The phenomenon was first experimentally observed in the rabbit hippocampus in the early 1970s, a brain region known to be critical for memory consolidation. The term “potentiation” refers to the increase in synaptic strength, while “long-term” denotes the effect’s sustained nature, lasting from hours to weeks. Its discovery provided the first compelling cellular model for memory storage.
Mechanism
The induction of LTP is heavily dependent on the activation of N-methyl-D-aspartate (NMDA) receptors on the postsynaptic membrane. High-frequency stimulation leads to a massive influx of calcium ions through these receptors, triggering a cascade of intracellular signaling events. This cascade ultimately results in the insertion of more alpha-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid (AMPA) receptors into the postsynaptic membrane, making the neuron more sensitive to future signals from the same presynaptic partner.
