Meaning

The JAK2-STAT5 Pathway is a crucial intracellular signaling cascade utilized by various peptide hormones and cytokines, most prominently Growth Hormone (GH) and prolactin, to rapidly transmit extracellular signals directly to the cell nucleus. This non-receptor tyrosine kinase pathway involves the Janus Kinase 2 (JAK2) and the Signal Transducer and Activator of Transcription 5 (STAT5), which together mediate the transcription of genes essential for cell proliferation, differentiation, and the powerful anabolic effects of GH. Its integrity is fundamental for normal growth, immune function, and metabolic homeostasis. Dysregulation, such as activating mutations in JAK2, is implicated in several hematological malignancies and endocrine disorders.
