Inflammation Mediators are a diverse group of biologically active molecules, including cytokines, chemokines, prostaglandins, and leukotrienes, that are released by cells in response to tissue injury, infection, or chronic stress. These chemical signals orchestrate the inflammatory response, a critical component of innate immunity, by regulating vascular permeability, recruiting immune cells, and initiating pain pathways. Clinically, chronic elevation or dysregulation of these mediators is central to the pathogenesis of numerous age-related diseases and hormonal imbalances.
Origin
The concept of chemical mediators of inflammation dates back to the early 20th century, with the identification of histamine and other vasoactive substances. The term itself is rooted in immunology and pathology, describing the molecules that “mediate” or carry out the actions of the inflammatory process. Understanding these mediators is essential for developing targeted anti-inflammatory therapies.
Mechanism
The mechanism begins when pattern recognition receptors on immune cells detect a threat, activating intracellular signaling cascades like the NF-κB pathway. This activation triggers the transcription and subsequent release of pro-inflammatory cytokines, such as Interleukin-6 (IL-6) and Tumor Necrosis Factor-alpha (TNF-α). These released mediators then bind to specific receptors on target cells, propagating the inflammatory signal and inducing systemic effects, including fever and the production of acute-phase proteins by the liver.
