It comprises the hypothalamus in the brain and the pituitary gland situated below it, which communicate via releasing and inhibiting hormones and direct neural connections. Origin
The term is derived from the anatomical structures involved—the hypothalamus (Greek hypo, under, and thalamos, inner chamber) and the pituitary gland (Latin pituita, phlegm, an older, incorrect term for its secretion)—linked by the concept of an axis to denote their functional relationship. The recognition of this hierarchical control structure emerged from mid-20th-century neuroendocrinology research. The HPA is often expanded to include a third target organ, such as the adrenal (HPA axis) or gonadal (HPG axis).
Mechanism
The hypothalamus synthesizes and releases neurohormones (e.g., TRH, GnRH, CRH) that travel through the hypophyseal portal system or direct nerve tracts to the pituitary gland. These signals either stimulate or inhibit the pituitary’s secretion of its own tropic hormones (e.g., TSH, LH, FSH, ACTH). These tropic hormones then travel through the general circulation to stimulate peripheral endocrine glands, which in turn release hormones that feed back to the hypothalamus and pituitary, completing the regulatory loop.
