Growth Hormone Insensitivity

Meaning

Growth Hormone Insensitivity (GHI) is an endocrine disorder characterized by a significantly reduced or entirely absent biological response to circulating growth hormone (GH), despite the presence of normal or even elevated levels of the hormone itself. This condition results from a defect in the signaling pathway downstream of the GH receptor, leading to a profound systemic deficiency in Insulin-like Growth Factor 1 (IGF-1) production. Clinically, GHI manifests as severe short stature, a key feature of Laron Syndrome, and is associated with distinct metabolic and body composition profiles. Understanding the molecular basis of this peripheral insensitivity is crucial for targeted therapeutic intervention in the somatotropic axis.
