Glucolipotoxicity is a pathogenic concept in endocrinology that describes the cellular dysfunction and damage induced by the chronic simultaneous exposure to elevated levels of both glucose and free fatty acids, or lipids. This combined metabolic stress is a central driver in the progression of insulin resistance and the decline of pancreatic beta-cell function, particularly in type 2 diabetes. The term highlights how the synergy of hyperglycemia and hyperlipidemia creates a toxic environment for sensitive metabolic tissues.
