Factor V Leiden is a specific genetic mutation in the gene for Factor V, a coagulation protein involved in the blood clotting cascade. This inherited condition results in a Factor V protein that is resistant to inactivation by activated Protein C, a natural anticoagulant, leading to a state of hypercoagulability. Individuals with this mutation have an increased, though variable, lifetime risk of developing venous thromboembolism (VTE), which includes deep vein thrombosis and pulmonary embolism. Clinical screening is often indicated in patients with a personal or family history of unexplained clotting events or prior to initiating certain hormonal therapies, such as oral contraceptives.
