Origin
This concept gained prominence with the recognition of environmental endocrine-disrupting chemicals, or EDCs, which were initially identified in environmental toxicology studies and later linked to human physiological alterations. The term is rooted in Greek, where exo- means “outside” and genous means “producing,” literally referring to an external source. Clinical endocrinology has since expanded this to include the effects of certain pharmaceuticals, phytoestrogens, and nutritional supplements.
Mechanism
The mechanism of action is often molecular mimicry, where the exogenous compound structurally resembles a natural hormone and binds to its receptor, acting as either an agonist to stimulate or an antagonist to block the normal cellular response. These agents can also interfere with key steroidogenic enzymes necessary for hormone synthesis, such as aromatase or 17-beta hydroxysteroid dehydrogenase. The resulting effect is a perturbation of the delicate neuroendocrine feedback loops that maintain homeostasis.
