The Estrone to Estradiol Ratio, often expressed as E1:E2, is a clinical metric comparing the circulating levels of the two primary estrogens, estrone (E1) and estradiol (E2). Estradiol is the most potent and biologically active estrogen, predominantly produced by the ovaries in premenopausal women, whereas estrone is a weaker estrogen that becomes the primary form after menopause, mainly produced in adipose tissue. A high E1:E2 ratio may suggest altered estrogen metabolism, potentially reflecting increased peripheral aromatization or reduced conversion of estrone to the more protective estradiol, which is a consideration in personalized hormonal risk assessment.
