Estrogen quinones are reactive, highly electrophilic metabolites formed during the oxidative breakdown of catechol estrogens, specifically the 4-hydroxyestrone and 4-hydroxyestradiol intermediates. These compounds are of significant clinical concern because they are unstable and can readily undergo redox cycling, generating reactive oxygen species that inflict oxidative stress on cellular components. Crucially, they possess the ability to covalently bind to DNA, forming depurinating DNA adducts that are considered initiating lesions in the development of certain hormone-sensitive cancers.
