Estrogen depletion refers to the clinical state characterized by a significant, sustained reduction in the circulating levels of biologically active estrogen hormones, primarily estradiol, below the physiological range required for optimal function. This hormonal deficit is most commonly observed during the menopausal transition, but it can also result from ovarian failure, certain medical treatments, or intense physical stress. The resulting lack of estrogenic signaling profoundly affects multiple organ systems, including the skeletal, cardiovascular, and urogenital systems.
