Dose-Limiting Toxicities, often abbreviated as DLTs, refer to adverse effects of a therapeutic agent that are severe enough to prevent the administration of further dose escalation in a clinical trial or require a reduction in the prescribed dose during routine clinical practice. These toxicities define the upper boundary of a drug’s safe dosage, indicating the maximum tolerable dose a patient can receive without experiencing unacceptable or life-threatening side effects.
