Origin
The term is a chemical and physiological descriptor, combining the name of the steroid molecule, DHEA, with the sulfate group attached to it. DHEA was first isolated from urine in the 1930s, and its sulfated form was later recognized as the primary circulating and storage form. The adrenal cortex is the main site of its production, reflecting the gland’s critical role in steroidogenesis.
Mechanism
The adrenal glands secrete DHEA, which is then rapidly converted to the more stable DHEA-Sulfate by the enzyme sulfotransferase. This sulfation makes the hormone more water-soluble, increasing its half-life in the circulation and allowing it to be transported efficiently to various tissues. Once in the target tissue, the sulfate group can be removed by steroid sulfatase, regenerating active DHEA which can then be metabolized into potent sex hormones like testosterone and estradiol, a process known as intracrinology.
