Meaning

De novo methylation refers to the enzymatic addition of methyl groups to previously unmethylated cytosine residues within DNA, primarily at CpG dinucleotides. This fundamental epigenetic process is catalyzed by specific DNA methyltransferases, notably DNMT3A and DNMT3B, which establish new methylation patterns distinct from the maintenance methylation carried out by DNMT1. This initial tagging of DNA sequences plays a critical role in establishing stable gene expression profiles across various cell types.
