Mechanism
The process begins intracellularly with the transcription and translation of procollagen polypeptide chains, followed by critical post-translational modifications like hydroxylation, which requires Vitamin C. These chains then assemble into a triple helix structure, procollagen, which is secreted into the extracellular space. Extracellularly, enzymes cleave the procollagen to form tropocollagen, which then spontaneously assembles into mature, cross-linked collagen fibrils. Estrogen and Growth Hormone significantly upregulate the activity of dermal and bone fibroblasts, thereby enhancing the rate of this entire biosynthetic pathway.
