Clomiphene, formally known as clomiphene citrate, is a synthetic, non-steroidal pharmaceutical agent classified as a Selective Estrogen Receptor Modulator (SERM) primarily used in reproductive endocrinology to stimulate the endogenous production of gonadotropins. Its clinical utility is focused on inducing ovulation in anovulatory women and stimulating the hypothalamic-pituitary-gonadal (HPG) axis in hypogonadal men to increase testosterone synthesis. The drug functions by strategically blocking the negative feedback effects of circulating estrogens at the level of the hypothalamus and pituitary gland, leading to an amplified pulsatile release of upstream regulatory hormones. It is a foundational therapeutic agent for non-exogenous modulation of the reproductive endocrine axis.
