The term combines Greek roots: an- meaning “without,” orexis meaning “appetite,” and genos meaning “producing,” thus describing something that “produces a lack of appetite.” The concept of a “feedback loop” originates from general systems theory, applied here to endocrinology to describe the continuous, self-regulating circuit of hormonal signals.
Mechanism
Following nutrient ingestion, various peripheral tissues, notably the gut and adipose tissue, release satiety hormones such as Peptide YY, Cholecystokinin, and Leptin. These gut peptide mediators and adipokines travel through the circulation to the arcuate nucleus of the hypothalamus in the brain. Upon reaching the hypothalamus, they activate Pro-opiomelanocortin (POMC) neurons, which then release anorexigenic neuropeptides like alpha-melanocyte-stimulating hormone. This central activation suppresses the orexigenic signals, resulting in a perceived reduction in hunger and the termination of a meal.
