Accelerated Blood Clearance, often abbreviated as the ABC phenomenon, is an immunological reaction observed in pharmacokinetics that significantly reduces the systemic circulation time of certain intravenously administered drug carriers. This phenomenon is a critical concern in the development and efficacy of nanomedicines, including those used for hormone delivery or targeting endocrine tissues. The rapid clearance compromises the intended therapeutic window and biodistribution profile of the encapsulated substance. It is characterized by a premature, rapid uptake of the drug carrier by the reticuloendothelial system, particularly in the liver and spleen.
